General description

Monoclonal Anti-CTGF (mouse IgG1 isotype) is derived from the hybridoma CTGF-14 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide. Connective tissue growth factor (CTGF) gene is mapped to human chromosome 6q23.2. It encodes a splice variant and also known as CCN family member 2 (CCN2). CTGF belongs to the family of regulatory proteins of the extracellular matrix (ECM) and is secreted by endothelial cells. It is expressed in the embryonic stage. The domain regions associated with the CTGF comprise four modules namely: Insulin-like growth factor binding proteins module, von willebrand factor type C repeat, thrombospondin type I repeat and cystine knot associated carboxy-terminal domain.

Specificity

Monoclonal Anti-CTGF recognizes human CTGF.

Immunogen

immunized with a synthetic peptide corresponding to an internal region of human CTGF (GeneID: 1490), conjugated to KLH. The corresponding sequence differs by a single amino acid in mouse and rat.

Application

Monoclonal Anti-CTGF antibody produced in mouse may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Connective tissue growth factor (CTGF) has been shown to play a role in developmental regulation of chondrogenesis, osteogenesis and angiogenesis, and in pathological processes, including fibrosis and tumorigenesis. CTGF may play an important role in the progression of malignant melanoma and the activation of wingless (WNT) signaling through low-density lipoprotein receptor-related protein 6 (LRP6). Certain polymorphisms in the CTGF gene have been linked with a higher incidence of systemic sclerosis.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage, freeze at -20oC in working aliquots. Repeated freezingand thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
