General description

Nestin (NES) is a class VI cytoplasmic protein which is expressed in the intermediate filaments. It has a molecular weight of 177.4kDa. The gene encoding it is localized on human chromosome 1q23.1 and has four exons.
Nestin is expressed during early stages of development in the central nervous system (CNS) and in myogenesis. Nestin is found in neurogenic progenitor cells (NPCs) and has been used as a marker for neural progenitor cells.

Immunogen

synthetic peptide corresponding to a sequence at the C-terminal region of human nestin, conjugated to KLH. The sequence is highly conserved (72% identity) in rat and mouse nestin.

Biochem/physiol Actions

Nestin (NES) functions as a modulator of proteins which are involved in cell growth. It also interacts with other proteins of the intermediate filament. NES has been studied as a marker of neural stem cells.
Nestin expression is down-regulated and replaced by the expression of other intermediate filament proteins such as neurofilament and Glial fibrillary acidic protein (GFAP). Nestin expression is re-induced in the adult organism during pathological conditions, such as formation of glial scar after injury to the central nervous system (CNS) and in certain types of CNS tumors. Nestin has been shown to play an important role in the phosphorylation-dependent disassembly of vimentin intermediate filaments during mitosis and is a potential target for Cyclin-dependent kinase 5 (cdk5) and p35 during development.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
