General description

Ras related protein consists of three isoforms such as Rab5a, Rab5b and Rab5c. Rab5a is encoded by the gene mapped to human chromosome 3p24.3. This gene codes for 23.66 kDa Rab5a protein.

Immunogen

synthetic peptide corresponding to amino acid residues 176-188 of human Rab5 with C-terminal added cysteine, conjugated to KLH.

Application

Anti-Rab5 antibody produced in rabbit has been used in western blotting and immunocytochemistry.

Biochem/physiol Actions

Rab proteins are members of small GTPases control the exocytosis and endocytosis and regulate vesicular transport. Rab5 regulates endocytic pathway in mammalian cells and an activator of autophagy. It mediates macroautophagy of cells expressing aggregates of mutant Huntingtin protein.
Rab5a is essential for epidermal growth factor (EGF)-mediated EGF receptor degradation. Overexpression of Rab5a is associated with the development of variety of cancers such as breast cancer, lung cancer, hepatocellular carcinoma (HCC) and ovarian cancer.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, and 15 mM sodium azide.
