General description

The gene ITGB3 (integrin subunit β 3) encodes the β subunit of integrins, which are transmembrane α/β heterodimers. There are 18 α subunits and 8 β subunits identified in mammals that associate with each other to form 24 different heterodimers. Based on the type of β3 subunit, the heterodimers can be grouped into subfamilies.

Immunogen

synthetic phosphopeptide derived from the region of human integrin β3 receptor that contains tyrosine 773. The sequence is conserved in human, mouse, and rat.

Biochem/physiol Actions

Integrins function in various cellular processes, such as tissue migration during embryogenesis, cellular adhesion, cancer metastases, and lymphocyte helper and killer cell functions. The β3 subunit is expressed along with αv and αIIb subunits in platelets to form the subfamilies αvβ3 and αIIbβ3. The receptor αvβ3 facilitates platelet adhesion to osteopontin and vitronectin in vitro. But, it is yet to be determined if it functions in platelet adhesion in vivo. The αvβ3 receptor functions as a receptor for fibrinogen, vWF, fibronectin, and vitronectin and participates in platelet aggregation. Any defects in the number or function of this receptor may result in the inhibition of platelet aggregation, leading to the bleeding disorder Glanzmann thrombasthenia.

The platelet integrin alpha(IIb)beta(3) is required for platelet aggregation. Like other integrins, alpha(IIb)beta(3) resides on cell surfaces in an equilibrium between inactive and active conformations. Recent experiments suggest that the shift between these conformations involves a global reorganization of the
