General description

Rho guanine nucleotide exchange factor 1 (ARHGEF1) is a member of a family of Rho-specific guanine nucleotide exchange factors. It contains a regulator of G protein signaling (rgRGS) domain near its amino-terminus. ARHGEF1 also has a tandem Dbl homology (DH)-Pleckstrin homology (PH) domain.

Application

Biochem/physiol Actions

Rho guanine nucleotide exchange factor 1(ARHGEF1) mediates dissociation of GDP and the binding of GTP. It binds specifically to the nucleotide-depleted state of the substrate. ARHGEF1 acts as a regulator of Rho and plays an important role in the activation of Ras-like proteins. Its amino terminal region helps it to function as an intermediate in the regulation of Rho proteins by G protiens G13 and G12. The GTPase activities of G12 and G13 are stimulated by it. Once G13 is activated by ARHGEF1, it catalyzes the nucleotide exchange on Rho. In arterial smooth muscle cells, it is responsible for angiotensin II-mediated activation of RhoA.
