General description

DPYSL3 (dihydropyrimidinase-like 3) belongs to collapsin response mediator proteins (CRMPs) family, which are phosphoproteins that reside in the cytoplasm. In humans, this family consists of five members. DPYSL3 has two isoforms due to alternate splicing. The long isoform consists of an extra domain in the N-terminal, the function and structure of which is not known. This family of proteins is highly expressed in the nervous system during developmental stages. This gene is localized to human chromosome 5q32. DPYSL3 is not found in adult brain, but is expressed in brain and spinal cord of fetus.

Application

Biochem/physiol Actions

DPYSL3 (dihydropyrimidinase-like 3) aids semaphorin/collapsin to induce collapse of the growth cone, regulates the differentiation of neurons and aids in axon guidance. It also plays an essential role in the development of the human nervous system. Its expression is induced in a certain subset of nervous system tumors and might act as an oncofetal neural antigen. This protein aids in the alignment of chromosomes by facilitating the bundling of F-actin, and therefore, mediates microtubule dynamics in mitosis. DPYSL3 suppresses metastasis in prostate cancer but facilitates the same in lung cancer. It is also linked with poor prognosis in neuroblastoma and pancreatic cancer. Polymorphism in this gene is also implicated in amyotrophic lateral sclerosis (ALS) in French population. In ischaemic stroke, it plays a role as a therapeutic target. DPYSL3 is involved in the tumorigenesis of pancreatic ductal adenocarcinoma, and has potential as a marker and therapeutic target for the same.
