Specificity

Clone 10C5 is a mouse monoclonal antibody that detects Tau. It targets an epitope within 52 amino acids in the proline-rich region (PR-1) of human Tau.

Immunogen

His-tagged, recombinant, human Tau isoform 1N3R aggregates produced with arachidonic acid.

Application

Quality Control Testing
Evaluated by Western Blotting with His-tagged, Human recombinant Tau, isoform 1N4R. Western Blotting Analysis: A 1:1,000 dilution of this antibody detected His-tagged, Human, recombinant Tau isoform 1N4R.

Tested Applications
Enzyme Immunoassay (ELISA): Serial dilutions from a representative lot detected Tau monomers or Tau aggregates induced with arachidonic acid in ELISA (Courtesy of Nicholas Kanaan, Ph.D., Michigan State University, USA).

Western Blotting Analysis: A representative lot detected recombinant, human Tau isoform 2N4R monomers and aggregates, 2N3R monomers and aggregates, and mouse Tau monomers. (Courtesy of Nicholas Kanaan, Ph.D., Michigan State University, USA).

Immunohistochemistry Applications: A representative lot detected Tau in Alzheimer′s diseased (AD) brain (superior temporal cortex, STG) tissue sections. Human disease sections had to be treated with phosphatase (dephosphorylates tau) to obtain reactivity suggesting tau in human AD is phosphorylated at its epitope. Target description

Microtubule-associated protein tau (UniProt: P10636; also known as Neurofibrillary tangle protein Paired helical filament-tau (PHF-tau)) is encoded by the MAPT (also known as MAPTL, MTBT1, TAU) gene (Gene ID: 4137) in human. Tau is a highly abundant protein found predominantly in the nervous system. Its primary role is to stabilize and support microtubule assembly, which are essential for maintaining the structure and function of nerve cells. It is present throughout the brain, particularly in regions involved in memory and cognition. MAPT gene consists of multiple exons that undergo alternative splicing, resulting in the generation of different isoforms that differ in the number and arrangement of repeats in the microtubule-binding domain. The expression of the MAPT transcripts varies and depends on the stage of neuronal maturation and neuron type. Aberrant aggregation and accumulation of tau protein have been linked to several neurodegenerative diseases collectively known as tauopathies. The most common tauopathy is Alzheimer′s disease, where hyperphosphorylated tau protein forms neurofibrillary tangles inside neurons. Other tauopathies include frontotemporal dementia, progressive supranuclear palsy, and cortico-basal degeneration. These diseases are characterized by cognitive decline, motor disturbances, and memory loss.

Physical form

Purified mouse monoclonal antibody IgG2b in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.

Reconstitution

1.0 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
