Target description

Kinesin-like protein KIF18A (UniProt: Q8NI77; also known as Marrow stromal KIF18A, MS-KIF18A) is encoded by the KIF18A (also known as OK/SW-cl.108) gene (Gene ID: 81930) in human. KIF18A is a widely expressed member of the kinesin superfamily of microtubule-associated molecular motors that hydrolyze ATP to produce force and movement along microtubules. Its kinesin motor domain is localized in amino acids 11-355. It is essential in aligning mitotic chromosomes at the spindle equator during cell division. It plays a role in chromosome congression by reducing the amplitude of pre-anaphase oscillations and slows poleward movement during anaphase, leading to suppression of chromosome movements. It preferentially concentrates on microtubules (MT) plus ends attached to a lagging kinetochore. It possesses both plus-end-directed motility and microtubule depolymerizing activity. It is reported to depolymerize longer microtubules more quickly than shorter ones. The depletion of KIF18A is reported to induce aberrantly long mitotic spindles and loss of tension across sister kinetochores, which results in the activation of the Mad2-dependent spindle-assembly checkpoint. Overexpression of KIF18A is reported in several cancers and is linked to poor overall survival. It is shown to accelerate the invasion and migration of head and neck squamous cell carcinoma.
