General description

Members of the F-box protein family, such as SCRAPPER, are characterized by an approximately 40-amino acid F-box motif. SCRAPPER is selectively expressed in the brain, broadly expressed within the mouse CNS, and is abundant at the presynaptic membrane. It has orthologs in C. elegans, D. melanogaster, and mammals which suggests that it might function as an important membrane-localized E3 ligase in various species. SCRAPPER is a major presynaptic E3 ubiquitin ligase that acts through RIM1a via degradation and the ubiquitin-proteasome-system (UPS) pathway to critically regulate synaptic transmission. This identifies protein degradation as a mechanism for holding synaptic communication in check.

This SCRAPPER antibody is validated for use in WB & IHC for the detection of the SCRAPPER protein.

Western Blot Analysis: 1 µg/mL from a representative lot detected SCRAPPER in 10 µg/mL of RAW264.7 cell lysate.

Immunohistochemistry Analysis: A 1:500 dilution from a representative lot detected SCRAPPER in human midbrain tissue.

Quality

Evaluated by Western Blot in human spleen tissue lysate.

Western Blot Analysis: 1 µg/mL of this antibody detected SCRAPPER in 10 µg/mL of human spleen tissue lysate.

Target description

~48 kDa observed. Uncharacterized bands may appear at ~52 kDa, ~58 kDa and ~70 kDa in some lysates.
