Specificity

Clone 3D13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Tyrosine-protein kinase receptor Axl (UFO). It targets an epitope within 23 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal, cytoplasmic domain of human Tyrosine-protein kinase receptor Axl (UFO).

Application

Quality Control Testing

Evaluated by Western Blotting in COS-7 and NIH3T3 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Axl (UFO) in COS-7 cell lysate and 1:1,000 dilution detected Axl (UFO) in NIH3T3 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Axl in U-251 MG cells.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected Axl (UFO) in one million U251 MG cells.

Affinity Binding Assay: A representative lot of this antibody bound Axl (UFO) peptide with a KD of 3.0 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Axl (UFO) in human skeletal muscle tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Tyrosine-protein kinase receptor UFO (UniProt: P30530; also known as EC:2.7.10.1, AXL oncogene) is encoded by the AXL (also known as UFO) gene (Gene ID: 558) in human. AXL is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 26-451), a transmembrane domain (aa 452-472), and a cytoplasmic domain (aa 473-894). It serves as receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to its ligand the growth arrest specific protein 6 (GAS6) via amino acids 26-92. Ligand binding at the cell surface induces dimerization and autophosphorylation of AXL that activates its kinase activity. Its protein kinase domain is localized in amino acids 536-807. Following its activation, AXL binds and induces tyrosine phosphorylation of PI3-kinase subunits PIK3R1, PIK3R2 and PIK3R3. Other known downstream substrate candidates for AXL are CBL, NCK2, SOCS1 and TNS2. Recruitment of GRB2 and PI3-kinase regulatory subunits by AXL also leads to the downstream activation of the Akt. AXL regulates several physiological processes including cell survival, cell proliferation, migration, and differentiation. The AXL/GAS6 signaling is associated with tumor growth, metastasis, invasion, epithelial-mesenchymal transition, angiogenesis, and drug resistance. Higher expression of AXL is reported in metastatic colon and breast cancer; however, its expression in normal colon tissue is weak. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
