Specificity

Clone 3I18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects DNA damage-inducible transcript 3 protein (DDIT3/GADD153).

His-tagged, full-length recombinant Human DNA damage-inducible transcript 3 protein (DDIT3/GADD153).

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cells nuclear extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected GADD153/DDIT3 in NIH3T3 cells nuclear extract.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GADD153/DDIT3 in lysate from C6 cells and in HeLa cells treated with Tunicamycin (5 g/mL; 6 h).

Enzyme Immunoassay Analysis (ELISA): Serial dilutions from a representative lot detected recombinant Human GADD153/DDIT3.

Affinity Binding Assay: A representative lot of this antibody bound GADD153 DDIT3 with a KD of 7.2 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

DNA damage-inducible transcript 3 protein (UniProt: P35638; also known as DDIT-3, C/EBP zeta, C/EBP-homologous protein, CHOP, C/EBP-homologous protein 10, CHOP-10, CCAAT/enhancer-binding protein homologous protein, Growth arrest and DNA damage-inducible protein GADD153) is encoded by the DDIT3 (also known as CHOP, CHOP10, GADD153) gene (Gene ID: 1649) in human. GADD153 is a multi-functional transcription factor that participates in endoplasmic reticulum (ER) stress response and induces cell cycle arrest and apoptosis in response to ER stress. It is localized in the cytoplasm under non-stressed conditions and translocates to nucleus under following ER stress. Its N-terminal region contains the transcriptional activation domain, and the C-terminal has basic-leucine zipper (bZIP) domain consisting of a basic amino-acid-rich DNA-binding region followed by a leucine zipper dimerization motif. It also contains two adjacent serine residues (79 and 82) that can serve as substrates of the p38 MAP kinase family. It is phosphorylated at serine residues by MAPK14, which enhances its transcriptional activation. However, phosphorylation of serine residues by casein kinase 2 (CK2) is reported to inhibit its transcriptional activation activity. GADD153 plays a regulatory role in the inflammatory response through the induction of caspase-11, which can induce the activation of caspase-1 (CASP1) leading to the activation of pro-IL-1β to mature IL-1β. It is reported to positively regulate the transcription of TRIB3, IL6, IL8, IL23, DR5, GADD34, PUMA, BIM, and ERO1L and negatively regulates the expression of Bcl-2, MYOD1. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
