Specificity

Clone 4-10-C9 is a ZooMAb® mouse recombinant monoclonal antibody that specifically detects LAG3. It targets an epitope within D3/D4 region from the extracellular domain.

Immunogen

Murine LAG-3-expressing mouse T-cell hybridoma.

Application

Quality Control Testing

Evaluated by Western Blotting with recombinant fragment corresponding to the extracellular domain of Mouse LAG3.

Western Blotting (WB) Analysis: A1:1,000 dilution of this antibody detected recombinant fragment corresponding to the extracellular domain of Mouse LAG3 fused to Fc portion of mouse IgG2a.

Tested Applications

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected LAG3 in Mouse splenocytes treated with Concanavalin A (2 g/mL; 3 days).

Affinity Binding Assay:: A representative lot of this antibody bound recombinant fragment corresponding to the extracellular domain of Mouse LAG3 with a KD of 2.8 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Lymphocyte activation gene 3 protein (UniProt: Q61790; LAG-3, Activation-induced cytidine deaminase-linked autoimmunity protein, Aida, CD223) is encoded by the Lag3 gene (Gene ID: 16768) in murine species. LAG3 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 24-442), a transmembrane domain (aa 443-463), and a cytoplasmic domain (aa 464-521). It contains three Ig-like C2-type domains and one Ig-like V-type domain. LAG3 is primarily expressed in activated CD4+ and CD8+ T-cells. Its expression is also observed in a subset of regulatory T cells. It serves as an inhibitory receptor on antigen activated T-cells and deliver inhibitory signals upon binding to ligands, such as Fibrinogen-like protein 1 (FGL1). It can negatively regulate the proliferation, activation, effector function and homeostasis of both CD8+ and CD4+ T-cells. Following

Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
