General description
Indole is the primary product of tryptophan breakdown by tryptophanase. Structurally, it is characterized by a benzene ring fused to a pyrrole ring, containing a five-membered nitrogen-containing ring. This simple structure forms the core of a vast array of natural and synthetic compounds with a wide range of biological activities. In bacteria, indole is a critical signaling molecule, produced as a metabolite during the breakdown of the amino acid tryptophan. It plays a pivotal role in bacterial behavior, influencing biofilm formation, spore germination, and the expression of virulence factors. In the context of metabolism in larger organisms, indole derivatives are integral to the functioning of the central nervous system, serving as precursors for the synthesis of neurotransmitters such as serotonin and melatonin. These compounds are consequential in regulating mood, appetite, sleep, and circadian rhythms. Principle
The indole assay kit is based on a modified version of Ehlrich′s and Kovac′s reagents, which reacts with indole to produce a colored compound at 565 nm. The intensity of this colored compound is directly proportional to the indole in the sample. Linear detection range from 3 to 100 μM indole in 96-well plate assay.
