Gene Information

General description

Thyrotropin receptor (UniProt: P16473; also known as Thyroid-stimulating hormone receptor, TSH-R) is encoded by the TSHR (also known as LGR3) gene (Gene ID: 7253) in human. TSH-R is a multi-pass membrane protein of the G-protein coupled receptor (GPCR) family that is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off to generate the mature form that contains four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. The extracellular domain is linked to a distal signal-specific domain the hinge region that is attached to a transmembrane domain consisting of extracellular and intracellular loops. It serves as a receptor the thyroid-stimulating hormone and TSH binding to this receptor induces thyrocyte growth and proliferation primarily by activating the adenylate cyclase signaling pathway. Clone CS-17 acts allosterically and serves as an inverse agonist for TSH-R constitutive activity. When administered in vivo it is shown to suppress thyroid function and reduces serum T4 levels in BALB/c mice. (Ref.: Chen, RR., et al. (2018). Thyroid. 28(77); 933-940; Chen, CR., et al. (2007). Endocrinology. 148(5); 2375-2382).

Specificity

Clone CS-17 is a mouse monoclonal antibody that detects human Thyrotropin receptor. It targets an epitope within the first extracellular domain from the N-terminal region.

Immunogen

Adenovirus expressing TSH receptor fragment (aa 22-289). adenovirus.

Application

Quality Control Testing

Evaluated by Flow Cytometry in TSH-R transfected HEK293 cells.

Flow Cytometry Analysis: 1 μg of this antibody detected TSH Receptor in one million TSH-R transfected HEK293 cells.

Tested Applications

Flow Cytometry: A representative lot detected TSH Receptor in Flow Cytometry applications (Chen, C.R., et al. (2007). Endocrinology. 148(5):2375-82; Chen, C.R., et al. (2009). Endocrinology. 150(7):3401-8; Chen, C.R., et al. (2018). Thyroid. 28(7):933-940).

Inhibition: A representative lot inhibited the constitutive activity of the TSH Receptor. (Furmaniak, J. et al. (2012). Autoimmun Highlights. 4(1):11-26).

Radioimmunoassay: A representative lot detected TSH Receptor in Radioimmunoassay applications (Chen, C.R., et al. (2007). Endocrinology. 148(5):2375-82).

Antagonist Function: A representative lot bloced TSH binding to the TSH Receptor. Endocrinology. 150(7):3401-8; Chen, C.R., et al. (2018). Thyroid. 28(7):933-940).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
Anti-TSH Receptor, clone CS-17, Cat. No. MABS2240, is a mouse monoclonal antibody that detects Thyroid-stimulating hormone receptor (TSH-R) and is tested for use in Flow Cytometry, Antagonist Function, Inhibition Assay, and Radioimmunoassay.

Physical form

Purified mouse monoclonal antibody IgG2a in PBS without azide.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
