General description

HuR , also known as Hu-antigen R, ELAV-like protein 1, and encoded by the gene name ELAVL1 and HUR, is a member of the RRM ELAV family. HuR is an RNA bindnig protein that contains 3 RNA-binding domains. It is known to binds AU-rich elements if mRNA. It regulates gene expression by destabilizing their mRNA. Recent studies have shown that HuR is upregulated in certain cancers such as breast and gastric cancers. HuR is also involved in embryonic stem cells (ESCs) differentiation. HuR preferentially binds mRNAs that are not methylated by N6-methyladenosine (m6A), stabilizing them, and thus promoting ESCs differentiation. Additionally, HuR has been shown to bind to poly-U elements and AU-rich elements (AREs) in the 3′-UTR of target mRNAs, as well as the AU-rich element in FOS and IL3/interleukin-3 mRNAs. Some of HuR’s targets include cFos, IL-3, and BRCA1 mRNAs. HuR is also involved in the regulation of VEFG-A and NFkB expression in certain cell types. HuR activation and accumulation are also linked to many energy signaling molcules such as that of the PI3K/Akt pathway and AMPK.
