General description

Human hormone-sensitive lipase (HSL) gene, containing 9 exons and spanning ~11kB, is mapped to chromosome 19q13.3. HSL is majorly expressed in adipose and steroidogenic tissues, but expressed at low levels in heart and skeletal muscle, macrophages, and islets. Functional HSL exists as a homodimer with high hydrolytic activity. HSL is characterized with a C-terminal catalytic domain, regulatory module and an N-terminal domain responsible for protein–protein and protein–lipid interactions.

The antiserum was produced against synthesized peptide derived from human HSL.

Biochem/physiol Actions

Hormone-sensitive lipase (HSL) is the major source of energy for most of the tissues. It plays a vital role in hydrolysis of triacylglycerol stored in adipose tissue and possibly steroidogenesis. HSL activity is regulated through reversible phosphorylation by catecholamines and insulin. HSL, also hydrolyzes cholesterol esters in the adrenals, ovaries, testes, and macrophages and plays an essential role in spermatogenesis. HSL mRNA expressed in islets might be implicated in glucose-induced insulin secretion. Abnormality in the protein expression results in obesity, atherosclerosis and type II diabetes.
