General description

IL3 (interleukin 3) is an immunomodulatory cytokine and a multilineage hematopoietic growth factor. It is predominantly produced by activated CD4+ T cells and mast cells. Recombinant mouse IL-3 is a 15.1 kDa protein containing 135 amino acid residues.

Biochem/physiol Actions

IL3 (interleukin 3) was originally known as multicolony-stimulating factor due to its capability to promote the development of a wide variety of hematopoietic progenitor cells from bone marrow, such as basophils, neutrophils, eosinophils, erythrocytes, megakaryocytes, mast cells, macrophages, and dendritic cells. In vitro this protein facilitates murine hematopoietic progenitor cell differentiation, and the survival and activation of mature myeloid cells. It promotes the release of histamine and mast cell growth, differentiation, and mediator release, and hence, is implicated in allergic inflammation. It exhibits bi-functionality during osteoclastogenesis where it stimulates the development of osteoclast progenitors but suppresses the osteoclastogenic process. Certain variants in this gene confer protection against malaria attack, and thus, this gene might play a role in the pathophysiology of falciparum malaria.

Interleukin (IL)-3, a multilineage hematopoietic growth factor, is implicated in the regulation of osteoclastogenesis. However, the role of IL-3 in osteoclastogenesis remains controversial; whereas early studies showed that IL-3 stimulates osteoclastogenesis, recent investigations demonstrated that IL-3 inhibits osteoclast formation.
