General description

Rho guanine nucleotide dissociation inhibitor I (RhoGDI), also known as Rho GDP dissociation inhibitor α (ARHGDIA) is encoded by the gene mapped to human chromosome 17q25.3-25-3. RhoGDI is the member of Rho/Rac protein family. The encoded protein consists of 204 amino acids and has a molecular mass of 23,421,000 Mr. In mammals, three RhoGDI isoforms exists: RhoGDIα, RhoGDIβ and RhoGDIγ. RhoGDIα is widely expressed. RhoGDIβ is mainly expressed in hematopoietic cells and RhoGDIγ is present in the brain, lung, kidney and testis.
Rho proteins are basically described as guanine nucleotide-binding proteins (GNBPs). RhoGDI (Rho guanine nucleotide dissociation inhibitor)/ARHGDIA (Rho guanine nucleotide dissociation inhibitor) has three subtypes namely- RhoGDIα (Rho GDP dissociation inhibitor α), RhoGDIβ (Rho GDP dissociation inhibitor β) and RhoGDIγ (Rho GDP dissociation inhibitor γ). RhoGDIα is expressed everywhere. It is located on human chromosome 17q25.3. Its protein has 204 amino acids and belongs to the Rho/Rac family.

Specificity

The antibody recognizes Rho-GDI and cross-reacts with D4-GDI.

Immunogen

GST fusion protein corresponding to full-length human Rho-GDI.

Application

Anti-Rho-GDI antibody produced in rabbit has been used in immunoblotting.
Anti-Rho-GDI has been used in western blotting.

Biochem/physiol Actions

Rho proteins control the actin and microtubule cytoskeleton. RhoGDIα is necessary for the spatial and temporal modulation of RhoGNBPs (guanine nucleotide-binding proteins). In HCC (hepatocellular carcinoma) patients, absence of ARHGDIA (Rho guanine nucleotide dissociation inhibitor) affects the prognosis and induces invasion and metastasis of HCC cells.

Physical form

Solution in 0.1 M Tris-glycine, pH 7.4, containing 0.15 M NaCl and 0.05% sodium azide.
