General description

Anti-Digoxigenin, Fab fragments are Fab fragments from polyclonal anti-digoxigenin antibodies. Anti-Digoxigenin Fab fragments are preferentially used for secondary detection using a light chain specific anti-sheep conjugate.
Digoxigenin is a hapten, useful in labeling nucleic acids and in detection systems.

Specificity

The polyclonal antibody from sheep is specific to digoxigenin and digoxin and shows no cross-reactivity with other steroids, such as human estrogens and androgens.

Application

Anti-Digoxigenin Fab fragments are preferentially used for secondary detection using a light chain specific anti-sheep conjugate.
Use Anti-Digoxigenin, Fab fragments for the detection of digoxigenin-labeled compounds using:
  • ELISA
  • Immunohistocytochemistry
  • In situ hybridization
  • Western blot
  • Flow cell construction.

Sequence

The number of amino groups in the Fab fragments is not known, because polyclonal antibodies are not sequenced. But as a rule of thumb full length antibodies contain about 80 amino groups on average.

Preparation Note

Working concentration: Working concentration of antibody depends on application and substrate. The following concentrations should be taken as a guideline:
  • ELISA: 2 to 4 μg/ml
  • Immunohistocytochemistry: 0.5 to 1 μg/ml
  • In situ hybridization: 0.2 to 0.4 μg/ml
  • Western blot: 0.5 to 2 μg/ml

Storage conditions (working solution): Diluted antibody is stable at 2 to 8 °C for 12 hours.
Always prepare freshly!

Reconstitution

Add 1 ml PBS to a final concentration of 1 mg/ml. Centrifuge reconstituted antibody for 5 minutes at 10,000 rpm prior to each use, and pipet the necessary amount carefully from the surface.

Analysis Note

Preparation: After immunization with digoxigenin, sheep IgGs are purified by ion exchange chromatography. Specific IgGs are isolated by immunosorption. Fab fragments obtained after papain digestion are purified by gel filtration and lyophilized.
