Specificity

Recognizes rat Acid Sensitive Ion Channel 1 (ASIC1). The immunogen shows no significant sequence homology with ASIC-beta or other ASICs.

Immunogen

A 20 amino acid peptide sequence near the extracellular N-terminus of rat ASIC1 (Chen et al. 1998; Waldmann et al. 1997; Garcia-Anoveros et al. 1997; Waldmann et al. 1996).

The immunogen sequence has 100% homology with human sodium channel 2 or BNaC2.

Application

Detect Acid Sensitive Ion Channel 1 using this Anti-Acid Sensitive Ion Channel 1 Antibody validated for use in ELISA & WB.

Western blot: 1-10 μg/mL using ECL.

Immunohistochemistry: not tested. It is recommended that the antibody be tried at 1-20 μg/mL on paraformaldehyde fixed tissue.

ELISA: 0.5-1 μg/mL using 1 μg/mL control peptide (Catalog number AG394) per well.

Optimal working dilutions must be determined by the end user.

Physical form

Affinity purified immunoglobulin. Liquid in phosphate buffered saline containing 0.1% BSA.

Storage and Stability

Maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Acid-sensing ion channels (ASICs) are voltage-insensitive cation channels that contribute to cellular excitability. We previously reported that ASIC1 in pulmonary artery smooth muscle cells (PASMC) contribute to pulmonary vasoreactivity and vascular remodeling during the development of chronic hypoxia (CH)-induced pulmonary
