Anti-LONP1, clone 1I18 ZooMAb, Cat. No. ZRB1314, is a recombinant rabbit monoclonal antibody that specifically detects mitochondrial Lon protease homolog (LONP1) and has been tested for use in Immunocytochemistry, and Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LONP1 in A431, HEK293, and L6 cell lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LONP1 in human small intestine and rat colon tissue sections.
Immunocytochemistry Analysis: A 1:10-100 dilution from a representative lot detected LONP1 in HeLa, A431, HUVEC, NIH 3T3 and MCF-7 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Lon protease homolog, mitochondrial (UniProt: P36776; also known as EC:3.4.21.53, LONHs, Lon protease-like protein, LONP, Mitochondrial ATP-dependent protease Lon, Serine protease 15) is encoded by the LONP1 (also known as PRSS15) gene (Gene ID: 9361) in human. LONP is a homohexameric, nuclear encoded, mitochondrial matrix protein that acts as an ATP-dependent serine protease. It is expressed ubiquitously and is present at high levels in the liver, brain, heart, skeletal muscle, placenta. It is synthesized with a transit peptide (aa 1-67) that helps in its import into the matrix where it is cleaved off to generate the mature form. It is organized in a ring with a central cavity and the ATP-binding and proteolytic domains (AP-domain) form a hexameric chamber. Its N-terminal domain (aa 124-368) is crucial for the overall structure of the protein, maintaining it in a conformation allowing its proper functioning. It is highly conserved throughout evolution and is shown to be crucial for the maintenance of mitochondrial homeostasis. LONP mediates the selective degradation of mutant and abnormal proteins in mitochondria. It also has a chaperone function in the assembly of inner membrane protein complexes. It also participates in the regulation of mitochondrial gene expression and in the maintenance of the integrity of the mitochondrial DNA. Its DNA and RNA binding is stimulated by substrate and inhibited by ATP binding. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
