ColorWheel^® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel^® antibody with any ColorWheel^® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel^® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. Leukocyte surface antigen CD47 (UniProt: A0A0D9R1W9; also known as Integrin-associated protein, CD47) is encoded by the CD47 gene (Gene ID: 103228373) in Monkey. CD47 is a multi-pass transmembrane glycoprotein that is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to produce the mature form. It is widely expressed on all hematopoietic cells and most other cell types. The ligands of CD47 include SIRP , thrombospondin-1 (TSP-1), and integrins including vβ3 and 2β1. It plays a role in vβ3-mediated cell adhesion and Ca2+ influx, as well as in platelet activation via interaction with thrombospondin-1. In addition, CD47 modulates peripheral mononuclear (PMN) neutrophil migration across endothelial and epithelial monolayers. The rate of fMLP-stimulated PMN migration is shown to be decreased by CD47 functional blockage upon neutralizing antibody treatment, which can be partially reversed by PI3K inhibition. Following fMLP stimulation and after PMN transmigration, secondary granules-derived CD47 molecules are shown to accumulate on plasma membrane, resulting in increased cell surface CD47 immunoreactivity. Overexpression of CD47 is reported on the surface of many types of cancer cells where it forms a signaling complex with signal-regulatory protein (SIRP ), enabling the escape of these cancer cells from macrophage-mediated phagocytosis. Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

재구성

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

저장 및 안정성

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
