일반 설명
Cortactin is an F-actin binding protein. It belongs to a highly conserved family of perimembrane signaling protein. Cortactin contains a unique multi-domain structure consisting of 6.5 tandem repeats of a 37-amino acid sequence required for direct binding to F-actin, followed by an α-helix region, a proline-rich region, and three tyrosine residues targeted by Src related kinases. The C-terminal region contains a Src-homology 3 (SH3) domain.
Cortactin is encoded by the CTTN gene on chromosome 11 in humans. It is expressed in most adherent cells and is a prominent substrate of protein-tyrosine kinase Src. The protein localizes within lamellipodia and at the cell surface. Cortactin is also called as EMS1.

synthetic peptide corresponding to the mid-region of human cortactin (amino acids 361-380) conjugated to KLH.
애플리케이션
Anti-Cortactin (KE-20) antibody produced in rabbit is suitable for indirect immunofluorescence at a dilution of 1:500 using mouse fibroblasts NIH3T3 cell line and western blotting at a dilution of 1:1,000 using cytosol extract of rat brain, whole extract of chicken embryo fibroblasts, or whole cell extract of the human epidermoid carcinoma A431 cell line.
생화학적/생리학적 작용
Cortactin is a cytoskeleton protein that facilitates the assembly of cortical actin. It is activated by phosphorylation of tyrosine kinases or serine/threonine kinases. It plays an important role in promoting lamellipodia formation, invadopodia formation, cell migration, and endocytosis. Cortactin regulates the interactions between components of adherens-type junctions and helps in organizing the cytoskeleton, cell adhesion structures of epithelia and carcinoma cells. During apoptosis, the encoded protein is degraded in a caspase-dependent manner. It acts as a regulator in invasion and migration of tumor cells. Overexpression of cortactin causes osteosarcoma carcinogenesis and along with FAK increases the risk of oral premalignant lesions.
물리적 형태

The release of late spermatids from the seminiferous epithelium requires the internalization of intercellular junctions by Sertoli cell specific structures called "tubulobulbar complexes" (TBCs). These large, endocytic devices likely evolved from classic clathrin-mediated-endocytosis (CME) machinery, but have several important morphological

Tyrosine phosphorylation evokes functional changes in a variety of ion channels. Modulation of the actin cytoskeleton also affects the function of some channels. Little is known about how these avenues of ion channel regulation may interact. We report that the

Invadopodia are dynamic protrusions in motile tumor cells whose function is to degrade extracellular matrix so that cells can enter into new environments. Invadopodia are specifically identified by microscopy as proteolytic invasive protrusions containing TKS5 and cortactin. The increasing complexity

The cortactin (CTTN) gene has been found, by transcriptomic profiling, to be overexpressed in pediatric osteosarcoma. The location of CTTN at 11q13 and the role of cortactin in cytoskeleton restructuring make CTTN of interest as a potential biomarker for osteosarcoma.
