Suitable for in vitro phosphorylation of proteins and peptides, phosphopeptide synthesis, characterization of phosphorylating sites, screening of activators and inhibitors, and mechanistic studies. The following substrates have been characterized by CK-II both in vitro and in vivo: nucleolin, RNA polymerase, p53, and SV 40 large T antigen.

Casein kinase II (CK2) is an active serine/threonine protein kinase that plays a pivotal role in cell growth and division. It is responsible for phosphorylating various nuclear proteins involved in DNA metabolism, transcription factors, and oncogenes, thus influencing their activity. It also regulates osteogenesis, adipogenesis, chondrogenesis, neuron differentiation, and immune response pathways. CK2 plays a crucial role in activating the Janus Kinase/signal transducer and activator of transcription (JAK/STAT) pathway, which contributes to cell survival, differentiation, and proliferation. Furthermore, CK2 is involved in activating the nuclear factor kappa B (NF-κB) pathway. It is highly expressed in different hematological malignancies, including acute lymphoblastic leukemia (ALL), acute myeloid leukemia (AML), chronic lymphocytic leukemia (CLL), and chronic myeloid leukemia (CML).
