General description

HiPrep Heparin FF 16/10 is prepacked with Heparin Sepharose 6 Fast FLow for preparative purification of proteins with affinity for heparin such as DNA-binding proteins, coagulation factors, lipoproteins, and protein synthesis factors.

Application

The column provides fast, preparative separations of Proteins and other biomolecules. Immobilized heparin has two main modes of interaction with Proteins. It can operate as an affinity ligand with affinity for a wide range of biomolecules, including coagulation factors and other plasma Proteins, lipoProteins, Protein synthesis factors, enzymes that act on nucleic acids, and steroid receptors. Heparin also functions as a high-capacity cation exchanger, due to its anionic sulfate groups. Gradient elution with salt is most commonly used in both cases. Heparin is coupled to Sepharose 6 Fast FLow with a chemically optimized linkage.

Heparin is a mucopolysaccharide consisting of a repeating dimer of α-L-dipyranuronic acid 2-sulfate, and 2-deoxy-2-sulfamino-α-D-glucopyranose 6-sulfate, with a molecular weight range of 5 000 to 30 000.
