Gene Information

General description

Immunoaffinity purified rabbit polyclonal antibody. Recognizes the ~53 kDa serotonin 2A (5-HT2A) receptor.
Recognizes the ~53 kDa serotonin 2A (5-HT2A) receptor.
This Anti-Serotonin 2A Receptor Rabbit pAb is validated for use in Frozen Sections, WB, Free Floating Sections, Paraffin Sections for the detection of Serotonin 2A Receptor.

Immunogen

Rat
a synthetic peptide corresponding to amino acids 22-41 of rat 5-HT2A receptor

Application

Frozen Sections (1:300-1:500)
Immunoblotting (1:500-1:1000)
Free Floating Sections (see application references)
Paraffin Sections (see application references; heat pre-treatment required)

Warning

Toxicity: Standard Handling (A)

Physical form

In 150 mM NaCl, 20 mM sodium phosphate, 1% BSA, pH 7.5.

Reconstitution

Following initial thaw, aliquot and freeze (-20°C).

Other Notes

Antibody specificity was examined in the rat amygdala, cortex, and hippocampus. Detection of primary antibody with ABC according to manufacturers directions. Low levels of glutaraldehyde (0.1%-0.3%) may be used in conjunction with paraformaldehyde for tissue perfusion fixation. Addition of intensifying reagents such as nickel ammonium sulfate to the chromogen solutions allows further dilution of the primary antibody to approximately 1:1000 or greater. For staining paraffin sections, heating in 10 mM citrate buffer is required (see application references). Antibody should be titrated for optimal results in individual systems.

The rodent hippocampus supports non-spatial object memory. Serotonin 5-HT2A receptors (5-HT2AR) are widely expressed throughout the hippocampus. We previously demonstrated that the activation of 5-HT2ARs enhanced the strength of object memory assessed 24 h after a limited (i.e., weak memory) training

Spaced training is robustly superior to massed training, which is a well-documented phenomenon in humans and animals. However, the mechanisms underlying the spacing effect still remain unclear. We have reported previously that spacing training exerts memory-enhancing effects by inhibiting forgetting
