Specificity

Reactive with human integrin alphaVbeta5. Does not recognize integrin alphaVbeta3 or alpha5beta1. The antibody binds to the alphaVbeta5 complex. The monoclonal antibody MAB2019Z has a Kd of 2.8 nM in Mn2+, 4.0 nM in Ca2+and 2.9 nM in buffer lacking cation. Thus, cations do not significantly affect the binding of the antibody to alphaVbeta5. Monoclonal antibody MAB2019Z does not block ligand binding to purified alphaVbeta5, nor does it block cell adhesion mediated through this receptor.

Immunogen

Generated against human integrin alphaVbeta5 purified from placental tissue and depleted of integrin alphaVbeta3.

Application

Anti-Integrin αVβ5 Antibody, clone 15F11, Azide Free is an antibody against Integrin αVβ5 for use in ELISA, FC, IP & IC.
Immunoprecipitation

Flow Cytometry: MG-63 cells have been harvested, blocked for 20 minutes at 4C in 20% normal human serum then washing in medium containing 10% FBS. 5X10E5 cells per tube were subsequently incubated with primary antibody for 30 min on ice, washed with medium, and incubated with fluorescent-conjugated secondary antibody.

Immunocytochemistry: 15μg/mL dilution for immunostaining of cells adhering to vitronectin-coated coverslips and fixed in 1% paraformaldehyde containing 0.5% Triton X-100 for 20 minutes. Incubation with MAB2019Z antibody for 30 minutes at 22C, followed by detection with a fluorescein-conjugated secondary antibody.
