General description

Specificity

Clone 2N13 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects APOA1.

Immunogen

His-tagged, recombinant fragment corresponding to 243 amino acids from human APOA1.

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected APOA1 in HepG2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected APOA1 in lysate from human liver tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected APOA1 in human liver tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected APOA1 in HepG2 cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected APOA1 in one million HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound APOA1 protein with at least ten thousand-fold (10,000X) higher than with non-specific GST control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Apolipoprotein A-I (UniProt: P02647; also known as Apo-AI, ApoA-I, Apolipoprotein A1) is encoded by the APOA1 gene (Gene ID: 335) in human. Apo A-1 is a major protein of plasma high density lipoprotein (HDL) and is also found in chylomicrons. It plays a major role in transporting cholesterol from peripheral tissues to the liver. In hepatocytes and enterocytes, APOA1 is localized in the endoplasmic reticulum (ER) during its synthesis. After post-translational modification, it is secreted into the extracellular space where it circulates in the blood bound to HDL particles. APOA1 is predominantly expressed in the liver and, also in small intestine. The protein is synthesized with a signal peptide (aa 1-18), which is proteolytically cleaved to generate the mature chain, which is cleaved into 2 chains: Proapolipoprotein A-I (aa 19-267) and a Truncated apolipoprotein A-I (aa 25-266). Apo A-1 also serves as a co-factor for lecithin cholesterolacyltransferase (LCAT) that is involved in the formation of cholesteryl esters. Elevated levels of oxidized Apo A-1 (on Methionine 110 and 136) have been linked to increased risk for coronary artery disease. Alterations in the APOA1 gene can lead to several lipid metabolism disorders. Mutations in this gene have also been linked to HDL deficiencies that lead to premature coronary artery disease and a reduction in cellular cholesterol efflux. Mutations in APOA1 gene have also shown to result in amyloidosis with extensive visceral amyloid deposit and in the pathogenesis of amyloid polyneuropathy-nephropathy Iowa type, also known as familial amyloid polyneuropathy type III. This ZooMAb® recombinant rabbit monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
