Specificity

Clone 2C11 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Protein Kinase C β (PKCβ) phosphorylated on serine 660.

Immunogen

KLH-conjugated linear peptide corresponding to 12 amino acids surrounding phosphoserine 660 from the C-terminal region of human Protein Kinase C β (PKCβ).

Application

Quality Control Testing

Evaluated by Western Blotting in lysate from NIH3T3 cells treated with phorbol 12-myristate 13-acetate (PMA).

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PKC β phosphorylated on serine 660 in lysate from NIH3T3 cells treated with phorbol 12-myristate 13-acetate (PMA; 400 ng/mL; 20 min).

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound p-PKC β (Ser660) peptide with a KD of 9.8 x 10-7 in an affinity binding assay.

Peptide Inhibition Assay: Target band detection in lysate from NIH3T3 cells treated with phorbol 12-myristate 13-acetate (PMA; 400 ng/mL; 20 min) was prevented by preblocking of a representative lot with the immunogen phosphopeptide, but not the corresponding non-phosphopeptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected phospho-PKCβ (Ser660) in serum starved NIH3T3 cells treated with phorbol 12-myristate 13-acetate (PMA; 400 ng/ml) for 20 min.).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Protein kinase C beta type (UniProt: P05771; also known as EC:2.7.11.13, PKC-B, PKC-beta) is encoded by the PRKCB (also known as PKCB, PRKCB1) gene (Gene ID: 5579) in human. PKCβ is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine kinase that is involved in multiple cellular processes, including the regulation of B-cell receptor, oxidative stress-induced apoptosis, insulin signaling, and endothelial cell proliferation. PKCβ is comprised of two splice variants, PKCβI and PKCβII, which are from the same gene and differ only in their last 50 amino acids. It binds three calcium ions per subunit and these ions are bound to the C2 domain (aa 158-275). Three specific sites: threonine 500 in the activation loop of the kinase domain; threonine 642 in the turn motif, and serine 661 in the hydrophobic region need to be phosphorylated for its full activation. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage.
