Specificity

Clone 3M19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Kinesin-like protein KIFC1. It targets an epitope within 22 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal region of human Kinesin-like protein KIFC1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell nuclear extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected KIFC1 in HeLa cell nuclear extract.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected KIFC1 in NIH3T3 cell nuclear extract.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected KIFC1 in Human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound KIFC1 peptide with a KD of 1.3 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected KIFC1 in U-251 MG cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Kinesin-like protein KIFC1 (UniProt: Q9BW19; also known as Kinesin-like protein 2, Kinesin-related protein HSET) is encoded by the KIFC1 (also known as HSET, KNSL2) gene (Gene ID: 3833) in human. KIFC1 is a minus end-directed motor protein, which is critical in centrosome clustering and is required for bipolar spindle formation. It is detected in the nucleus, cytoplasm, and early endosomes. It is shown to be associated with nucleus during interphase, centrosomes in early and spindle in later mitosis. Its conserved kinesin motor domain is localized in amino acids 310-663 and contains the microtubule-binding site and a catalytic region for the hydrolysis of ATP. Its tail domain can specifically recognize and bind to the membranous organelles, vesicles, and -tubulin. It is reported to transport various cargos along the microtubule to the minus ends. Deletion of KIFC1 gene leads to defects in metaphase mitotic spindle assembly, and results in chromosome structural abnormalities. Overexpression of KIFC1 is reported in patients with hepatocellular carcinoma and is linked to disease progression and poor prognosis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

30 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
