Specificity

Clone 1C2 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects C-terminal-binding protein 2 (CTBP2). It targets an epitope within 18 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the C-terminal region of human C-terminal-binding protein 2 (CTBP2).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CTBP2 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CTBP2 in C6 and NIH3T3 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CTBP2 in A431, HeLa, HUVEC, and NIH3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound CTBP-2 peptide with a KD of 4.0 x 10-6 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CTBP2 in human gallbladder tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

C-terminal-binding protein 2 (UniProt: P56545; also known as CtBP2) is encoded by the CTBP2 gene (Gene ID: 1488) in human. CTBP1 and CTBP2 are transcriptional co-repressors that modulate the activity of a large number of transcriptional repressors via the recruitment of chromatin modifiers. Although CTBP1 and CTBP2 show high degree of homology, their activity is differentially regulated, particularly through the control of their subcellular localization. It is only the CTBP2 that has a nuclear localization signal at its N-terminus. CTBP2 is a member of the D-isomer specific 2-hydroxyacid dehydrogenase family that is ubiquitous in its distribution with highest levels observed in heart, skeletal muscle, and pancreas. It can exist as a homodimer or can form heterodimer with CTBP1. The repressor activity of CTBP2 is reported to be reciprocally regulated by NADH and acyl-CoA. It represses FoxO1-mediated gluconeogenesis directly and sterol regulatory element-binding protein 1 (SREBP1)-mediated lipogenesis indirectly. Obese subjects exhibit defective CTBP2 activity in their liver. Deletion of CTBP2 is shown to promote hepatic gluconeogenesis and enhance steatohepatitis. It is also involved in brown adipose tissue differentiation. A strict positive correlation between CTBP2 and T-cell lymphoma invasion and metastasis 1 (Tiam1) protein expression has been reported. Tiam1 serves as a downstream target of CTBP2. Knockdown of CTBP2 is reported to reduce Tiam1 levels resulting in lower invasion and migration. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.03 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
