Specificity

Clone 1H21 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detect Profilin-1. It targets an epitope within 15 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human Profilin-1.

Application

Quality Control Testing

Evaluated by Western Blotting in COS-7 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Profilin-1 in COS-7 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Profilin-1 in lysates from HeLa, C2C12, and PC12 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Profilin-1 in Human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Profilin-1 in HeLa cells .

Affinity Binding Assay: A representative lot of this antibody bound Profilin-1 peptide with a KD of 9.5 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Profilin-1 (UniProt: P07737; also known as Epididymis tissue protein Li 184a, Profilin I) is encoded by the PFN1 gene (Gene ID: 5216) in human. Profilin 1 is a ubiquitous small actin-binding protein that is expressed in all eukaryotes. It promotes actin polymerization and plays a vital role in many cell functions, including membrane trafficking, endocytosis, cell cycle, motility, proliferation, cell survival, transcription, stemness, and autophagy. It enhances ADP-to-ATP exchange on G-actin and is capable of adding ATP-bound G-actin to the barbed ends of growing filaments. Phosphorylation of profilin 1 at serine 138 is reported to reduce its affinity for G-actin and block its interaction with HTT, reducing its ability to inhibit HTT aggregation. Profilin 1 is reported to be essential for retention and metabolism of hematopoietic stem cells in mouse bone marrow. It also participates in regulation of DNA replication. In unperturbed cells, it increases DNA replication initiation and accelerates fork progression by binding and stimulating the pyridoxal 5 -phosphate (PLP)-containing nucleosome remodeler SNF2H. Under replication stress, profilin1/SNF2H increases fork stalling and collaborates with fork reversal enzymes to enable the over-resection of unprotected forks. Mutants forms of profilin 1 form insoluble aggregates that disrupt cytoskeletal structure, and increase ubiquitin and p62/SQSTM levels in motor neurons. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
