Specificity

Clone 1B3 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Syntaxin 17 (STX17). It targets an epitope within 20 amino acids from the N-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal, cytoplasmic domain of human Syntaxin17 (STX17).

Application

Quality Control Testing

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected STX17 in A431 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected STX17 in lysates from HepG2 cells, mouse testis tissue, and C2C12 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected STX17 in human kidney tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected STX17 in A431 and HeLa cells..

Target description

Syntaxin-17 (UniProt: P56962; also known as STX17) is encoded by the STX17 gene (Gene ID: 55014) in human. STX17 is a multi-pass membrane protein with two cytoplasmic domains (aa 2-228 and 276-302), two helical domains (aa 229-249 and 255-275), and one lumenal domain (aa 250-254). STX17 is as an autophagosomal soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) that has a hairpin-type structure mediated by two transmembrane domains, each containing glycine zipper motifs. This structure contributes to its specific localization to completed autophagosomes. Mutations in the glycine residues on the glycine zipper-like motifs is shown to impair its localization to autophagosomes. Although STX17 is shown to localize to the endoplasmic reticulum and mitochondria under nutrient-rich conditions, it translocates to autophagosomes under condition of starvation. It is reported to interact with SNAP29 that is recruited to autophagosomes upon starvation, and VAMP8, the lysosomal SNARE. Depletion of either SNAP29 or VAMP8 is shown to block autophagic flux. STX17 also plays a role in the early secretory pathway where it may maintain the architecture of the endoplasmic reticulum-Golgi intermediate compartment and regulates transport between them. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
