Specificity

Clone 1G13 is a Rabbit recombinant monoclonal antibody that specifically detects AF4/FMR2 family member 2 (AFF2). It targets an epitope within 160 amino acids from the C-terminal half.

Immunogen

His-tagged recombinant fragment corresponding to 160 amino acids from the C-terminal half of human AF4/FMR2 family member 2 (AFF2).

Application

Quality Control Testing

Evaluated by Western Blotting in Neuro2A cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected AFF2 in Neuro2A cell lysate.

Tested applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected AFF2 in mouse brain tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound human recombinant AFF2 with a KD of 8.6 x 10-9 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

AF4/FMR2 family member 2 (UniProt: P51816; also known as Fragile X E mental retardation syndrome protein, Fragile X mental retardation 2 protein, FMR2P, Protein FMR-2, Protein Ox19) is encoded by the AFF2 (also known as FMR2, OX19) gene (Gene ID: 2334) in human. In mammals four different homologs of Drosophila Lilli protein have been described and are named as AFF1, AFF2, AFF3, and AFF4. AFF1 and 4 are part of super elongation complex (SEC) and are required to release polymerase II. AFF2 and 3 are key subunits of the SEC-like complexes SEC-L2 and SEC-L3, respectively. They are required for transcription elongation in some genes. AFF2 is RNA-binding protein that is involved in alternative splicing regulation through an interaction with G-quartet RNA structure. It associates with mRNA via one of the three RNA-binding domains. AFF2 is mainly expressed in the brain and is most abundant in hippocampus and amygdala. Some expression is also observed in placenta and lung. It localizes with splicing factor SC35 in nuclear speckles where splicing factors are concentrated, assembled, and modified. It is reported to regulates the transcription and toxicity of expanded GGGGCC (G4C2) repeat-containing C9ORF72 allele in Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). CRISPR-Cas9-mediated loss of AFF2 is shown to reduce levels of C9ORF72 RNA, RNA foci, and DPR proteins in neurons derived from iPSCs of patients with C9ORF72-ALS/FTD. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
