Specificity

Clone 2G13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects CD11b. It targets an epitope within 20 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the extracellular domain of human CD11b (Integrin alpha-M).

Application

Quality Control Testing

Evaluated by Western Blotting in Human brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD11b in Human brain tissue lysate.

Tested applications

Affinity Binding Assay: A representative lot of this antibody bound CD11b with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD11b in K562 and THP-1 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD11b in human bone marrow tissue sections.

Anti-CD11b, clone 2G13 ZooMAb®, Cat. No. ZRB1278, is a recombinant Rabbit monoclonal antibody that specifically targets CD11b and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Integrin alpha-M (UniProt: P11215; also known as CD11 antigen-like family member B, CR-3 alpha chain, Cell surface glycoprotein MAC-1 subunit alpha, Leukocyte adhesion receptor MO1, Neutrophil adherence receptor, CD11b) is encoded by the ITGAM (also known as CD11B, CR3A) gene (Gene ID: 3684) in human. Integrins are a/b heterodimeric adhesion receptors that couple the extracellular matrix on other cells with the contractile cytoskeleton to transduce mechanochemical signals across the plasma membrane. CD11b is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-16), which is subsequently cleaved off to generate the mature form that that contains a long extracellular domain (aa 17-1104), a transmembrane domain (aa 1105-1128), and a short cytoplasmic domain (aa 1129-1152). It is predominantly expressed in monocytes and granulocytes. It is usually found as a heterodimer of a/b subunits and associates with Integrin b2 (ITGB2). CD11b/Integrin b2 has been implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement-coated particles and pathogens. The major ligand recognition site of CD11b/integrin b2 has been mapped to the A-domain in the CD11b subunit (CD11bA). CD11b/integrin b2 also serves as a receptor for fibrinogen, factor X and ICAM1. It recognizes P1 and P2 peptides of fibrinogen gamma chain. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
