Specificity

Clone 3B17 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Arginase-1. It targets an epitope within 20 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal region of human Arginase-1.

Application

Quality Control Testing

Evaluated by Western Blotting in Human Liver tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Arginase-1 in Human Liver tissue lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Arginase-1in human liver tissue sections.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected Arginase-1 in one million HepG2 cells.

Affinity Binding Assay; A representative lot of this antibody bound Arginase-1 peptides with a KD of 1.3 x 10-7 in an affinity binding assay.

Anti-Arginase-1, clone 3B17 ZooMAb®, Cat. No. ZRB1088, is a recombinant Rabbit monoclonal antibody that targets Arginase-1 and is tested for use in Affinity Binding Assay, Flow Cytometry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Arginase-1 (UniProt: P05089; also known as EC:3.5.3.1, Liver-type arginase, Type I arginase) is encoded by the ARG1 gene (Gene ID: 383) in human. Arginase-1 is a manganese-containing liver enzyme that participates in urea cycle and converts L-arginine to urea and L-ornithine. It also functions to maintain L-arginine homeostasis in non-hepatic tissues where nitric oxide synthase (NOS) and arginase compete for the available intracellular arginine. Arginase-1 is also Involved in an antimicrobial effector pathway in polymorphonuclear granulocytes (PMN). Upon PMN cell death, it is liberated from the phagolysosome and depletes arginine in the microenvironment leading to suppressed T cell and natural killer (NK) cell proliferation and cytokine secretion. Although expression of arginase-1 is commonly associated with inflammation, fibrosis, and wound healing involving increased L-proline, polyamine, and Th2 cytokine production, macrophage-specific arginase-1 is reported to act as an inhibitor of inflammation and fibrosis following infection with the Th2-inducing pathogens. In murine species, arginase is induced in myeloid cells mainly by Th2 cytokines and inflammatory agents and participates in a variety of inflammatory diseases by down-regulation of nitric oxide synthesis, induction of fibrosis and tissue regeneration. Mutations in ARG1 gene are associated with Aargininemia that is characterized by elevated levels of arginine in the blood and cerebrospinal fluid, developmental delays, seizures, and mental retardation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
