Gene Information

human ... NTF3(4908)

General description

NTF3 (neurotrophin 3) is a neurotrophic factor belonging to the NGF (nerve growth factor) family. This protein is composed of 257 amino acids, with a prepro region of 138 amino acids and the mature sequence of 119 residues.

Biochem/physiol Actions

NT3 functions on two structurally unrelated receptor types- the p75 neurotrophin receptor (p75NTR) and the receptor tyrosine kinase (TRK). It promotes the metastatic invasion of brain by breast cancer cells, by promoting the EMT (epithelial-mesenchymal transition) transition of breast cancer cells and suppressing the immune response in brain. This protein is responsible for the indction and survival of neurite outgrowth from a subset of neural crest (NC) and placode-derived neurons.
Recombinant human NT-3 is a noncovalently linked homodimer, of two 13.6kDa polypeptide monomers (240 total amino acid residues).

Physical form

Lyophilized with no additives.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.

The development and maintenance of the vertebrate nervous system depends upon neuronal survival proteins known as neurotrophic factors. Nerve growth factor (NGF) remains the best characterized neurotrophic molecule. Brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3) are two recently cloned neurotrophic
