General description

Hybrisol® Hybridization Solutions are pre-made, ready-to-use solutions optimized for sensitive single and multiple gene copy detection using membrane-based hybridization techniques. They are designed to give maximum signal and clean background for hybridizations performed on nylon and nitrocellulose membranes. Hybrisol Solution contains 10% Dextran Sulfate, 1% SDS, 50% Formamide, 6X SSC, Sheared DNA, and Modified Denhardt′s Solution.

Application

All
Before using, preheat at 37°C to 45°C for 10 - 20 minutes to dissolve precipitants and mix thoroughly. Use 1 ml for every 15 cm2 membrane for prehybridization and hybridization. Prehybridize for 60 minutes at 42°C to 45°C. Hybridize overnight at 45°C to 52°C. Prehybridization and hybridization temperatures may have to be determined empirically. Refer to the Nomograph located under the "Protocols" link at S4040 product specific page at www.chemicon.com for hybridization temperatures with various stringencies guidelines.

Liquid Hybrisol I Solution contains 10% Dextran Sulfate, 1% SDS, 50% Formamide, 6X SSC, Sheared DNA, and Modified

Denhardt′s Solution.

Storage and Stability

Store at room temperature tightly sealed when not in use. Solution can be stored refrigerated however it should be warmed to at least room temperature prior to use dilution for best results. Precipitates may form when the solution is cooled.
