Gene Information

human ... NAMPT(10135)

General description

Nicotinamide phosphoribosyltransferase (NAMPT) is a member of the NAPRTase family. Its primary role is to catalyze the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, an intermediate in the biosynthesis of NAD. NAMPT is localized in the cytoplasm and expressed in large amounts in bone marrow, liver tissue, and muscle tissues. NAMPT inhibits neutrophil apoptosis in experimental inflammation and clinical sepsis.

Immunogen

Recombinant protein corresponding to mouse Nampt.

Application

Anti-Nampt antibody, clone 14A.5 is an antibody against Nampt for use in western blotting, IHC, inhibition & ELISA.
Western Blotting Analysis: 0.1 µg/mL from a representative lot detected Nampt in Anti-CD3epsilon stimulated mouse spleen cell lysate (Prof. Oberdan Leo, Universite Libre de Bruxelles.).

Western Blotting Analysis: A representative lot from an independent laboratory detected Nampt in mouse spleen tissue lysate (Ronveaux, A., et al. (2002). Eur J Immunol. 32(11):3225-3234.).

Inhibition Analysis: A representative lot from an independent laboratory was inhibited by a synthetic peptide corresponding to murine Nampt in total liver extracts (Ronveaux, A., et al. (2002). Eur J Immunol. 32(11):3225-3234.).

Immunohistochemistry Analysis: A 1:100 dilution from a representative lot detected Nampt in CD11c positive mouse spleen sections (Prof. Oberdan Leo, Universite Libre de Bruxelles.).

ELISA Analysis: A representative lot from an independent laboratory detected recombinant Nampt peptides (Ronveaux, A., et al. (2002). Eur J Immunol. 32(11):3225-3234.).

Quality

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected Nampt in 10 µg of Raji cell lysate.

The murine homologue of the previously identified human "pre-B-cell colony-enhancing factor" (PBEF) gene coding for a putative cytokine has been identified by screening a subtractive library enriched in genes expressed in activated T lymphocytes. Unlike most cytokine genes known to
