General description

Brevican core protein (BCAN), also known as Brain-enriched hyaluronan-binding protein (BEHAB) or Chondroitin sulfate proteoglycan 7 (CSPG7), may play a role in the terminally differentiating and the adult nervous system during postnatal development. It may stabilize interactions between hyaluronan (HA) and brain proteoglycans. Two isoforms exist, with isoform 1 (~99 kDa) being highly expressed from birth through 8 years of age and down-regulated by 20 years of age to low levels that are maintained in the normal adult cortex; and isoform 2 (~72 kDa) being expressed at uniformly low levels throughout development.

Immunogen

Recombinant protein corresponding to rat Brevican.

Application

Anti-Brevican, clone N294A/6 is an antibody targeting the Brevican protein, validated for use in WB & IHC.

Western Blot Analysis: 2 µg/mL from a representative lot detected Brevican in 10 µg of human brain tissue lysate.

Immunohistochemistry Analysis: A 1:500 dilution from a representative lot detected Brevican in rat midbrain, rat thalamus, and rat cerebellum tissues.

Western Blot Analysis: A representative lot detected Brevican in rat brain tissue lysate and in WT, but not in Brevican knockout mouse brain tissue lysate.

Quality

Evaluated by Western Blot in rat brain tissue lysate.

Western Blot Analysis: 2 µg/mL of this antibody detected Brevican in 10 µg of rat brain tissue lysate.

Target description

~140 kDa observed. The calculated molecular weight of this protein is 96 kDa A second isoform is known to exist at 69 kDa This protein may be observed at a higher molecular weight due to glycosylation. A proteolyticaly cleaved fragment of Brevican may be observed at ~50 kDa in some tissue lysates.

Evidence indicate that the brain-specific protein, brevican, is proteolytically cleaved during neurodegeneration, hence positioning fragments of brevican as potential blood biomarkers of neurodegenerative diseases, such as dementia. We aimed to develop two assays capable of detecting the brevican N-terminal (N-Brev)
