General description

Ribonuclease ZC3H12A (UniProt Q5D1E7; also known as MCP-induced protein 1, Regnase-1, Zinc finger CCCH domain-containing protein 12A) is encoded by the Zc3h12a (also known as Mcpip, Mcpip1) gene (Gene ID 230738) in murine species. Regnase-1 is an endonuclease that is involved in post-transcriptional gene regulation by cleaving target mRNAs or the terminal loop of pre-microRNAs. Regnase-1 negatively regulates Il6, Il12b, Il2, Icos, Ox40, Ctla4, cRel mRNAs, as well as its own mRNA (Zc3h12a). In T cells, regnase-1 and an RNA-binding protein roquin act together to inhibit T helper (TH) cell differentiation by negatively regulating mRNAs encoding the TH17 cell-promoting factors IL-6, ICOS, c-Rel, IRF4, IκBNS and IκBζ. This cooperation requires RNA-binding activity of roquin and the endonuclease activity of regnase-1. Upon T-cell receptor (TCR) activation following antigen binding, however, both roquin and regnase-1 are cleaved and inactivated by the paracaspase MALT1, allowing TH17 differentiation to proceed.

Specificity

Clone 15D11 detected regnase-1 in wild-type, but not Zc3h12a-/-, mouse embryonic fibroblasts/MEFs (Jeltsch, K.M., et al. (2014). Nat. Immunol. 15(11):1079-1089).

This Anti-Regnase-1/Zc3h12a Antibody, clone 15D11 is validated for use in Western Blotting for the detection of Regnase-1/Zc3h12a.
Western Blotting Analysis: A representative lot detected regnase-1 in lysates from wild-type, but not Zc3h12a-/-, mouse embryonic fibroblasts/MEFs (Jeltsch, K.M., et al. (2014). Nat. Immunol. 15(11):1079-1089).

Quality

Evaluated by Western Blotting in MEF1 cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected Regnase-1/Zc3h12a in 10 µg of MEF1 cell lysate.
