General description

Zinc finger and BTB domain-containing protein 7A (UniProt O95365; also known as Factor binding IST protein 1, Factor that binds to inducer of short transcripts protein 1, FBI-1, HIV-1 1st-binding protein 1, Leukemia/lymphoma-related factor, POK erythroid myeloid ontogenic factor, Pokemon, TIP21, POZ and Krueppel erythroid myeloid ontogenic factor, TTF-I-interacting peptide 21, Zinc finger and BTB domain-containing protein 7A, Zinc finger protein 857A) is encoded by the ZBTB7A (also known as FBI1, LRF, ZBTB7, ZNF857A) gene (Gene ID 51341) in human. Pokemon/ZBTB7A belongs to the family of POK (POZ/BTB and Krüppel) transcription factors that play important roles in cellular differentiation and oncogenesis. Pokemon/ZBTB7A overexpression is reported in cancers of various origins, including non–small-cell lung cancer (NSCLC), breast cancer, ovarian cancer, as well as in non-Hodgkin lymphoma, where it is found to directly repress the expression of the tumor suppressor ARF. On the other hand, loss of pokemon/ZBTB7A expression is also found to promote the progression of prostate tumorigenesis among a subset of human advanced prostate cancers by bypassing Pten loss–induced cellular senescence (PICS) as a result of Sox9-dependent oncogenic pathways activation.

Specificity

Clone 13E9 detected pokemon/ZBTB7A expression in wild-type, but not Zbtb7-/-, mouse embryonic fibroblasts by Western blotting and showed strong and specific nuclear staining only in Zbtb7+/+ fetal liver (FL) cells, but not in Zbtb7-/- FL cells (Maeda, T., et al. (2005). Nature. 433(7023):278-285).

Application

Anti-ZBTB7A Antibody, clone 13E9 is an antibody against ZBTB7A for use in Western Blotting, Immunohistochemistry (Paraffin).
Western Blotting Analysis: 2.0 µg/mL from a representative lot detected ZBTB7A in 10 µg of HepG2 and K562 cell lysates.
Western Blotting Analysis: A representative lot detected much lower pokemon/ZBTB7A exprerssion levels in 6 human prostate cell lines when compared with the non-tumourigenic RWPE-1 prostate epithelial cell line (Wang, G., et al. (2013). Nat. Genet. 45(7):739-746).
Western Blotting Analysis: A representative lot detected a downregulated pokemon/ZBTB7A exprerssion in human DU145 prostate cancer cells transfected with miR-106b or miR-93, but not miR-25 (Wang, G., et al. (2013). Nat. Genet. 45(7):739-746).
Western Blotting Analysis: A representative lot detected pokemon/ZBTB7A expression in prostates from wild-type mice, but not transgenic mice with prostate-specific Zbtb7a gene-knockout (Wang, G., et al. (2013). Nat. Genet. 45(7):739-746).
Western Blotting Analysis: A representative lot detected pokemon/ZBTB7A expression in wild-type mouse thymocytes and mEFs, but not Zbtb7-/- mEFs. Clone13E9 also detected the overexpressed pokemon levels in thymic lymphomas developed among lckEµ-Pokemon transgenic mice (Maeda, T., et al. (2005). Nature. 433(7023):278-285).
Immunohistochemistry Analysis: A representative lot detected a positive correlation between the pokemon/ZBTB7A immunoreactivity and that of PTEN in formalin-fixed, paraffin-embedded human prostate cancer tissues (Wang, G., et al. (2013). Nat. Genet. 45(7):739-746).
Immunohistochemistry Analysis: A representative lot detected nuclear pokemon/ZBTB7A immunoreactivity in human (thymus and tonsil) and mouse (thymus and spleen) lymphoid organs using paraffin-embedded tissue sections (Maeda, T., et al. (2005). Nature. 433(7023):278-285).
Immunohistochemistry Analysis: A representative lot detected specific nuclear pokemon/ZBTB7A immunoreactivity only in Zbtb7+/+ fetal liver (FL) cells, but not in Zbtb7-/- FL cells (Maeda, T., et al. (2005). Nature. 433(7023):278-285).
Immunohistochemistry Analysis: A representative lot detected pokemon/ZBTB7A immunoreactivity among subsets of human T-cell lymphomas, diffuse large B-cell lymphoma (DLBCL), and follicular lymphoma (FL) samples, while little pokemon immunoreactivity was seen in normal human thymic CD3+ cells (Maeda, T., et al. (2005). Nature. 433(7023):278-285).

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: 1.0 µg/mL of this antibody detected ZBTB7A in 10 µg of HeLa cell lysate.

Target description

~70 kDa observed. Target band appears larger than the calculated molecular weight of 61.4 kDa
