General description

Sulf-2 is also important in neurological situations including myelination and glial cell differentiation and GDNF activity. Sulf-2 activity is also found in the kidney and important in glomerular basement membrane development and glomerular function. Finally, Sulf-2 is involved in heparan sulfate metabolism. Sulf-2 removes sulfate moieties on heparan sulfate proteoglycans which in turn modulate the heparin binding growth factor signaling activity of the cell. Sulf-2 also impacts extracellular matrix interactions and it is important during development and growth, as well as Wnt signaling and VEGF production by endothelial cells during vascularization. In disease, Sulf-2 expression in cancer is upregulated and inhibition of Sulf-2 limits cancer progression and showed increased apoptosis within tumors, thus Sulf-2 activation is being examined as a potential anti-cancer therapy.

Application

This Anti-Sulf-2 Antibody, clone 2B4 is validated for use in Western Blotting and Immunohistochemistry and ELISA for the detection of Sulf-2.
Western Blot Analysis: 1 µg/mL from a representative lot of this antibody detected Sulf-2 in transfected HEK293 cell lysates (Data courtesy of Dr. Mark Singer).

Western Blot Analysis: A representative lot of this antibody detected Sulf-2 in transfected HEK293 cell lysates, but not in non-transfected HEK293 cell lysates (Lemjabbar-Alaoui, H., et al., (2010) Oncogene 29:635–646).

Immunohistochemistry Analysis: A representative lot of this antibody detected Sulf-2 in Human lung and squamouse cell carcinoma tissue (Lemjabbar-Alaoui, H., et al., (2010) Oncogene 29:635–646).

ELISA: A representative lot of this antibody detected Sulf-2, but not Sulf-1 by ELISA (Lemjabbar-Alaoui, H., et al., (2010) Oncogene 29:635–646).

Quality

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: 2 µg/mL of this antibody detected Sulf-2 in 10 µg of A549 cell lysate.

Target description

~125 & 50 kDa observed.
Sulf-2 is synthesized as a ~125 kDa pre-protein and is subsequently cleaved into ~75 kDa and ~50 kDa fragments, which are held together by disulfide bonds in the mature protein.
