General description

HU-B27 (HLA- Com-B27) is involved in the presentation of foreign antigens to the immune system. HLA B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta 2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells.

The previously assigned protein identifier P03989 has been merged into P01889. Full details can be found on the UniProt database.

Specificity

This reagent binds strongly to the HLA-B27 antigen. It also binds weakly to the HLAB7 antigen. This formulation demonstrates less cross reactivity to HLA-B7 than Chemicon products, MAB1285 and MAB1285F.

Application

Detect HU-B27d using this Anti-HU-B27 Antibody, FITC conjugated validated for use in FC, IC, IF & WB.
Flow cytometry and Immunofluorescence: use 10 mL direct from the vial per 100 mL of whole blood, or 1 x 106 peripheral blood mononuclear cells (PBMC) in 100 mL buffer.

Flow cytometric analysis of lymphocyte staining for HLA-B27 gave the following results:
Subjects/Fluorescence Intensity
Heterozygous HLA-B27+/Strong
Homozygous HLA-B7+/Weak
Heterozygous HLA-B7+/Weak - negative
Non B27, Non B7/Negative

This reagent is useful in the investigation of HLA-B27 in diseases such as ankylosing spondylitis.

SUGGESTED USAGE:
Flow cytometry and immunofluorescence - use 10 µl direct from the vial per 100 µl of whole blood, or 1 x 10E6 peripheral blood mononuclear cells (PBMC) in 100 µl buffer.

Use Control IgG2a-FITC (MABC002F) as an isotype control.

Please note that this reagent should be used in a single-colour single-step assay only.

Evaluated by Western Blot on Jurkat lysates.

Western Blotting Analysis:
1:500 dilution of this antibody detected Com-B27 on 10 µg of Jurkat lysates.

Physical form

The reagent is supplied in 1.0 mL phosphate-buffered saline, pH 7.4, containing 0.2% bovine serum albumin and 0.1% sodium azide. Purified from mouse ascitic fluid. The characteristics of each lot are tested by gel electrophoresis and flow cytometry.

Storage and Stability

Stable for 6 months at 2-8ºC in the dark from date of receipt.
