General description

Cyclic AMP-dependent transcription factor ATF-4 (ATF4) is a basic leucine-zipper (bZip) transcription factor, which regulates amino acid metabolism, DNA damage repair, chromatin remodeling, and apoptosis in response to cellular and ER stress. ATF4 works with various proteins, such as C/EBP homology protein (CHOP), aspargine synthetase (ASNS), and cAMP response element (CRE) among others to mediate cellular stress. ATF4 also regulates glucose homeostasis by suppressing beta-cell proliferation and insulin production. Furthermore, ATF4 targets the histone demethylase JMJD3 to alter chromatin structure and enhance gene transcription in response to amino acid deprivation.

Quality

Evaluated by Western Blot in untreated and HisOH treated HepG2 cell lysate.

Western Blot Analysis: A 1:2,000 dilution from a representative lot detected ATF4 in 10 µg of HisOH treated HepG2 cell lysate (HisOH was used to induce cellular stress mediated eIF2a phosphorylation, allowing for subsequent detection of ATF4. For enhanced performance, end-users may try higher dilutions of this antibody. For better results, 1% SDS was used during the secondary antibody incubation step.)

Target description

~48 kDa observed. The calculated molecular weight is 39 kDa; however, this protein may be observed at ~48 kDa in human tissue lysates and ~45 kDa in mouse cell lysates due to post-translational modification. Uncharacterized bands may be observed at ~57 kDa, ~59 kDa, ~75 kDa, and ~100 kDa in some cell lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
