Gene Information

human ... KMT2D(8085)

General description

MLL2, also known as Histone-lysine N-methyltransferase 2D, or Lysine N-methyltransferase 2D or ALL1-related protein, or Myeloid/lymphoid or mixed-lineage leukemia protein 2, and encoded by the gene KMT2D/ALR/MLL2/MLL4, is a histone methyltransferase that methylates Lys-4 of histone H3 to form H3K4me. H3K4me represents a specific tag for epigenetic transcriptional activation and for instance in estrogen receptor function where MLL2 acts as coactivator for estrogen receptor by being recruited by ESR1, thereby activating transcription. MLL2 is also a component of the ASCOM/Mll2/3 complex which plays an important role during development particularly in lymphomagenesis. MLL2 is localized to the nucleus. MLL2 is expressed in most embryonic and adult tissues. Defects in MLL2 are associated with Kabuki syndrome, a congenital mental retardation syndrome with additional features, including postnatal dwarfism, facial and bone deformities. Defects are also associated with medulloblastoma a highly malignant primary brain tumor.

HOXA transcript at the distal tip (HOTTIP), a long noncoding RNA, is upregulated in pancreatic ductal adenocarcinoma (PDAC), but the HOTTIP-mediated oncogenic pathway is not fully understood. We identified canonical HOTTIP-HOXA13 targets, CYP26B1, CLIC5, CHI3L1 and UCP2-responsible for cell growth

MLL2, an epigenetic regulator in mammalian cells, mediates histone 3 lysine 4 tri-methylation (H3K4me3) through the formation of a multiprotein complex. MLL2 shares a high degree of structural similarity with MLL, which is frequently disrupted in leukemias via chromosomal translocations.
