General description

The protein HEC1 (highly expressed in cancer 1) is an essential component of the vital NDC80 kinetochore-associated protein complex. The NDC80 complex is required for chromosome segregation and spindle checkpoint activity. HEC1 is essential for proper kinetochore integrity and the organization of stable microtubule binding sites in the outer plate of the kinetochore. HEC1 is expressed by all dividing cells and is particularly high in cancers. HEC1 localizes to kinetochores from late prophase to anaphase, specifically at the outer plate of the kinetochore. HEC1 is normally phosphorylated during its active phase at multiple sites and the phosphorylation appears to tune kinetochore-microtubule binding affinity during mitosis. Moreover, recent research using small molecule inhibitors to HEC1′s kinase, Nek2, demonstrate that phosphorylation is required for proper HEC1 function.

Immunocytochemistry Analysis: 1.0 µg/mL from a representative lot detected Hec1 in HeLa cells.
