General description

Purified mouse monoclonal antibody. Recognizes the ~170 kDa MDR1 isoform of P-glycoprotein.
Recognizes the ~170 kDa MDR1 isoform of P-glycoprotein. Does not cross-react with the MDR3 P-glycoprotein.
This Anti-P-Glycoprotein Mouse mAb (C494) is validated for use in Paraffin Sections, FC, Frozen Sections, Immunoblotting, ICC, IP for the detection of P-Glycoprotein.

Immunogen

Epitope: gene-specific internal cellular epitope present only on the MDR1 isoform of P-glycoprotein
Hamster and Human
SDS-solubilized plasma membranes of a multidrug resistant (MDR) Chinese hamster ovary (CHO) cell line and a human leukemic cell line

Application

Paraffin Sections (5-10 µg/ml, see comments)

Flow Cytometry (see comments)

Frozen Sections (5-10 µg/ml, see comments)

Immunoblotting (see comments)

Immunocytochemistry (see application references)

Immunoprecipitation (see comments)

Packaging

Please refer to vial label for lot-specific concentration.

Warning

Toxicity: Standard Handling (A)

Physical form

In PBS, 1% BSA.

Reconstitution

Following initial thaw, aliquot and freeze (-20°C).

Analysis Note

Positive Control
Cell lines or frozen tissue, such as human liver or colon.

Other Notes

Does not cross-react with MDR3 P-glycoprotein. Also reported to work for flow cytometry, immunoblotting, and immunoprecipitation. For immunocytochemistry, frozen or paraffin-embedded material may be used. Permeabilization of the cell membrane is required to expose the epitope recognized by this antibody. Variables associated with assay conditions will dictate the proper working dilutions.

Salmonella enterica serotype Typhimurium is a food-borne pathogen that also selectively grows in tumours and functionally decreases P-glycoprotein (P-gp), a multidrug resistance transporter. Here we report that the Salmonella type III secretion effector, SipA, is responsible for P-gp modulation through
