General description

PEMT (phosphatidylethanolamine N-methyltransferase) is an an important enzyme for the synthesis of membrane phospholipids. PEMT converts phosphatidylethanolamine to phosphatidyl-choline by sequential methylation in the liver. The protein localizes to the endoplasmic reticulum and mitochondria-associated membranes. The gene is within the Smith-Magenis syndrome region on chromosome 17. Inhibition of hepatic PEMT activity has been associated with Alzheimer’s disease and schizophrenia.

Specificity

Other homologies: Mouse (86% sequence homology) and Rat (73% sequence homology).
This antibody recognizes the C-terminal region of human PEMT in Jurkat cell lysates. The antibody easily detected PEMT in Jurkat and L6 lysates by Western blotting, but expression was weak or not detected in A431, HeLa, HEK293, PC12, or NIH/3T3 cell lysates using very dilute antibody (0.25 μg/mL, data not shown). Positive signals in immunocytochemistry using higher concentrations of antibody (2.5 μg/mL) detected PEMT in these cell lines, suggesting differential expression levels of this protein in various cell lines.

The immunogen was a KLH-conjugated synthetic peptide corresponding to the C-terminal region of human PEMT.

Immunocytochemistry:
HeLa, A431, and NIH/3T3 (previous page) cells incubated with anti-PEMT (C-terminus) stained positively on nuclei and endoplasmic reticulum (ER).

Storage and Stability

Stable for 6 months at 2-8°C from date of receipt. For long term storage, store at -20ºC. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance

Phosphatidylethanolamine is converted to phosphatidylcholine in mammalian liver by the enzyme phosphatidylethanolamine N-methyltransferase (PEMT). A form of the enzyme (PEMT2) has been isolated from rat liver, the cDNA cloned and expressed and the murine gene has been characterized and disrupted.
