General description

Histone H4 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail H4 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. Acetylation of histone H4 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs).

Specificity

Expected to cross-react with Tetrahymena and Eukaryote.
Other species not tested.
Recognizes hyperacetylated histone H4, Mr 10 kDa (SDS-PAGE); tri- and tetra-acetylated histone H4 on acid-urea gels. May cross-react with other acetylated proteins.

Immunogen

KLH-conjugated, synthetic peptide
(AGGAcKGGAcKGGAcKGGAcKGGAcKGGC) related to amino acids 2 to 19 of Tetrahymena thermophila histone H4.

Application

Chromatin Immunoprecipitation:
Reported to immunoprecipitate transcriptionally active chromatin containing acetylated histone H4. (Perry, C.A., 1993)

Immunocytochemistry:
Reported to exclusively stain Tetrahymena macronuclei1 and chromatin in HeLa nuclei. (Perry, C.A., 1993)

Western Blot Analysis:
1:1000 dilution of this lot detected HYPERACETYLATED HIS H4 on 10 μg of HepG2 lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
For maximum recovery of product, centrifuge the vial prior to removing the cap.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variabillity in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
